#exons
🧵 1/8 Have you ever wondered how information embedded within exons regulates gene expression in plants? 🌱🧬 Check out our latest work, where we explore how exonic enhancers (EEs) shape gene expression in plants! www.biorxiv.org/content/10.6...

the process of building an app is like the process of building a protein: there are the useful problem solving bits - exons - and then there are a whooole bunch more bits - introns - related to navigating whatever framework(s) you're using to make the exons possible

I’ve been studying gene regulation in primates for over 20 years, and using RNA-seq since 2008 to compare expression across species. We thought we were being careful. We used curated orthologous exons to minimize alignment bias and focused on biologically meaningful comparisons.

We were wrong.

The target wordcount for Exons was 100,000. Something nice and easy to submit to publishers.

Current wordcount? 101,221.

Is it finished, Anesha?

No. No it is not.

Es wird mal wieder Zeit für ein bisschen Molekularbiologie. Heute mit dem Thema "Splicing" 🧑‍🔬 🧬

Many conserved exons in the heart and brain utilize weak 5 ′ splice sites, yet they are accurately spliced. But how? We show that splicing fidelity is actively enforced through a QKI-U6 checkpoint at the U1→U6 handover in essential cardiac genes during organogenesis.
www.biorxiv.org/content/10.1...
QKI ensures splicing fidelity during cardiogenesis by engaging the U6 tri-snRNP to activate splicing at weak 5ʹ splice sites
During organogenesis, precise pre-mRNA splicing is essential to assemble tissue architecture. Many developmentally essential exons bear weak 5'splice sites (5'SS) yet are spliced with high precision, ...

Recommendation: One of the web serials I've been following for the past month is an apocalyptic TG virus story set in an dystopian near-future.

Playing with #vizitig again to prepare for the 1.0 release. This is the SNAP25 human gene expressed in 3 SRA datasets linked to cancers.
Exons have one color each, and exon junctions are in yellow.
The rest is new/noise.

How can a splicing factor bind to thousands of exons but regulate only a few of them? In our new preprint, we show that SRSF6 resolves this paradox through two distinct binding modes. @lab-nmr.bsky.social & @julianvonehr.bsky.social. Check it out:

I’m thrilled to share that my PhD work has been just published in Cell. After a long and bumpy ride, we uncovered the core function of nuclear speckles -splicing of GC-levelled exons- and traced the evolution of this gene architecture and condensates themselves to amniotes.

📣 Attention #RNA #Splicing lovers! How is splicing of exons coordinated??🤔 ➡️ Exon Junction Complex (EJC) coordinates inclusion of blocks of neighboring exons!😯 Cool work by Alexandra Bergfort @karlaneugebauer.bsky.social lab - check it out! Check out our new preprint on thousands of novel recursive exons in the human genome (if you don't know about recursive exons, they are pretty cool), led by PhD student David Bass @davidb256.bsky.social : www.biorxiv.org/content/10.6...
Large-scale analysis of transcript data reveals thousands of recursive splicing events in human introns
Recursive splicing (RS) is a process in which an intron is removed from a nascent RNA molecule in two or more splicing events rather than one. We introduce a novel approach for detecting recursive spl...

1\ Excited to finally share some of my PhD work: OpenSplice!

We measured how >590,000 mutations affect exon inclusion across 600 human exons, creating a resource to study splicing regulation, benchmark variant effect predictors, and support variant interpretation.

A gene is a segment of a DNA molecule with multiple regions: a promoter, exons (the bits that dictate amino acid sequence in a protein), introns, & terminator regions.

mRNA is a short-lived intermediate of exon segments of DNA — it is a TRANSCRIPT of a piece of the gene.
