#extrasinusoidal
This week in #Science #Immunology: Lukas Amann, Marco Prinz, & colleagues at @uni-freiburg.de report that #extrasinusoidal #macrophages in the #mouse #DuraMater represent a distinct subset of #CNS -associated macrophages whose role is shaped by #autoimmune #neuroinflammation! #neuroimmunology

For context, a 'extrasinusoidal' macrophages are macs NOT near sagittal (between the cerebral hemispheres) & transverse sinuses (sit between the cerebral hemispheres & cerebellum) - nice diagram to show this from their Fig S1 👇

🤩 paper in on extrasinusoidal macrophages in 🐭 dura from Marco Prinz's group led by Lukas Amann @uni-freiburg.de - this is a VERY comprehensive paper, lots to ❤️ but their role in phagocytosing dying neurophils at both the onset & peak of EAE neuroinflammation is quite interesting 🧪 #ImmunoSky 🧠

Our new study on Dura Mater macrophages regulating #brain #immunity at the interface between the brain and periphery - congrats Lukas Amann 💪💪💪

Extrasinusoidal dural macrophages were distinct from leptomeningeal and CNS parenchymal macrophages in terms of their origin, expansion kinetics, and transcriptional profiles. During autoimmune neuroinflammation, extrasinusoidal dural macrophages performed efferocytosis of apoptotic granulocytes.

Although the loss of Tln1 strongly impeded the formation of cellular protrusions by extrasinusoidal dural macrophages, they were still efficiently sampling soluble dextran, which is taken up via receptor-mediated endocytosis, for example through binding to CD206
