🧪🧬New preprint We present cryo-EM structures of reconstituted CTCF–nucleosome complexes, showing CTCF dimerization drives nucleosome oligomerization into defined higher-order assemblies. Disrupting CTCF–CTCF interfaces in mESCs reduces looping and impairs differentiation. tinyurl.com/CTCF-nucleos...

In our IWS1 story, we also visualize the histone reader LEDGF PWWP domain on a transcribed downstream nucleosome. Because the PWWP requires both DNA gyres for binding, it binds to the promoter-distal side of the nucleosome during transcription through the nucleosome.

Very nice new paper from Paggi, Long, Zhang using physical modeling of RCMC data to reconstruct physical structures of euchromatin at the nucleosome scale:

Euchromatin forms condensed domains and E-P interactions largely occur on the surface!

Nucleosome aficionados! Our new review "Nucleosome spacing across cell types, diseases, and ages" is out in NAR: academic.oup.com/nar/article/...

A huge effort to pull together what we’ve learned about nucleosome spacing in many systems. Enjoy!

Our collaborative paper with the Arndt lab is out today: doi.org/10.1093/nar/... (originally bioRxiv: www.biorxiv.org/content/10.1...). Here we asked the question of how CHD1 localizes to chromatin.
A direct interaction between the Chd1 CHCT domain and Rtf1 controls Chd1 distribution and nucleosome positioning on active genes
Abstract. The nucleosome remodeler Chd1 is required for the re-establishment of nucleosome positioning in the wake of transcription elongation by RNA Polym

BTS as histones in a nucleosome. BTS as a dance your PhD challenge.
The #BTS DNA choreography involves some deep biology where they dance a nucleosome. It's been shown that DNA (arms) tighten, loosen, and twist around histones (the guy's bodies) in cells which serves a packaging role and also affects which portions of the DNA can be easily read.
