Antigen-specific regulatory T cells eliminate cognate CD4 effector T cells through perforin, granzyme B, and Bim
Antigen-specific regulatory T cells eliminate cognate CD4 effector T cells through perforin, granzyme B, and Bim
Ye et al. describe a synthetic-lethality-analogous killing model. Shared peptide-MHC-II co-activation enables Treg perforin-granzyme B cytotoxicity; responder Bim confers susceptibility in vitro and in mouse airway inflammation.

The immune protein perforin can kill tumor cells without major granzymes. Cell experiments point to an iron-dependent death pathway, but clinical applications remain uncertain. doi.org/hcqqzv
Immune system protein is a cancer killer on its own, new study shows
For decades, scientists have known that a protein made by T cells and natural killer (NK) cells in the immune system, called perforin, can kill tumor cells by punching a hole in the cell wall and injecting proteases called granzymes, which kill the cell.

Bell et al. discovered a previously unrecognized, granzyme-independent function of perforin (Prf1) in CTLs and NK cells that showed that Prf1 can directly kill tumor cells via ferroptosis.  @umich.edu

Perforin triggers iron-induced mitochondrial damage to drive ferroptosis and anti-tumor immunity
Perforin is best known for delivering granzymes into tumor cells, but this study shows it has a second, independent killing function: it drives iron uptake and mitochondrial iron overload that trigger...

How do killer immune cells destroy tumor cells without granzymes?

Just out in Cell: Prf1 directly induces tumoral ferroptosis by driving endosomal-mitochondrial contacts that cause mitochondrial iron accumulation and dysfunction, in a Gzm-independent manner.

Ev-Marie Schuster generated some stunning microscopy images showing that CAR and TCR synapses can form simultaneously with two different target cells – and that the stronger receptor effectively steals perforin from the weaker one!

Why does this matter?

Forest exposure is associated with higher natural killer cell activity. These are the immune cells that patrol your body looking for infected and abnormal cells.

It's also linked with higher levels of perforin, the spear-like protein NK cells use to punch holes in compromised cells.
