Disentangling the Promotional Roles of Interfacial and Alloyed Sites in PtCr Catalysts for CO2 Hydrogenation
Promotional effects are ubiquitous across catalytic processes involving supported nanoparticles, where additional elements, known as promoters, significantly enhance the catalytic performances (activity, selectivity, and/or stability) of nanoparticles. However, the inherent complexity of catalytic materials, comprising multiple species located both in the bulk and at the surface, makes it difficult to pinpoint the role of promoters at the molecular level. In this study, we disentangle the effect of alloying and interfacial sites in a low-temperature reverse water–gas shift (RWGS) reaction, a key process in the chemical industry, by precisely constructing catalysts featuring narrowly dispersed alloyed PtCr nanoparticles with or without Cr(III) interfacial sites. Notably, we show that a catalyst containing exclusively PtCr alloys, PtCr@SiO2, displays a substantial increase in catalytic activity compared to monometallic Pt@SiO2, while having additional Cr(III) interfacial sites (PtCr-Crint@SiO2) further improves the catalyst performance. In situ spectroscopic results reveal that the PtCr alloy facilitates a redox reaction pathway, whereas the presence of Cr(III) interfacial sites greatly facilitates CO2 adsorption and opens an additional formate-mediated pathway, further accelerating the reaction. These findings highlight the power of well-defined model systems in elucidating the promotional effects at the molecular level.

I visited the factory ~two weeks ago and, serendipitously, got to show John how we use nano-DESI with PTCR MSn for membrane proteins in tissue. His description of our ions as “precious” (a very nice way to say absurdly low ion counts…) will stick with me!

Overall, PTCR proves once again to be a useful gas-phase reaction that enables us to simplify our spectra to see the complexity of glycoproteoforms. We are excited to keep exploring this technology to better understand glycoproteoforom landscapes and use that to connect glyco regulation to function.

I work in sport governance with a PTCR within the Coaching Association of Canada.

When the Olympics decided to allow professional athletes to compete in the Olympics, the line got murky.

The reality is that most Olympic athletes and coaches are not wealthy and need support to compete. Cheers!

Improved Protein Identification in Native Ambient Mass Spectrometry by the Integration of Proton Transfer Charge Reduction and Higher-Energy Collision Dissociation (PTCR-HCD MS 3 )
AbstractHere, we demonstrate PTCR-HCD MS3 as a tool for improving protein identification in native ambient mass spectrometry (NAMS). NAMS involves the direct analysis of tissue sections with no or little sample preparation and no (solution-phase) separation postsampling. NAMS samples are therefore highly complex, comprising proteins, salts, and other biomolecules, and NAMS spectra are highly congested. PTCR MS has emerged as a useful strategy in top-down proteomics. Most commonly, it is applied as an adjunct MS2 step, providing molecular weight information to support sequence information provided by collision-based MS2 or it is applied as an MS3 step to simplify collision, photon, or electron-based MS2 by reducing m/z overlap of product ions. In this work, we show that the selection of charge-reduced PTCR product ions for subsequent HCD reduces the incidence of chimeric fragmentation spectra and improves confidence in protein identification in NAMS samples from mouse spinal cord, rat kidney, and cell-line-derived xenograft tissue. As well as removing interference from overlapping protein species, we show that PTCR-HCD MS3 also removes interference from overlapping lipid species which can similarly affect confident protein identification. The approach also allows identification of proteins that are masked at the MS1 level but are revealed following PTCR.

We found that collecting DIA-PTCR spectra with high resolving powers (Rp) provided significant benefits. Prior work had focused on low Rp data collection (which can limit glycoproteoform assignments), so we describe our method as high resolving power (hRp)-DIA-PTCR to help emphasize this benefit.
