Among the sarbecovirus subgenus, SARS-CoV-2 is the only one so far discovered with a furin cleavage site at the S1-S2 junction.

It is also the only sarbecovirus to have caused a pandemic.

Further, several common cold CoVs have furin cleavage sites and so do other betacoronaviruses (the genus).

Could we eventually have a vaccine that neutralizes many variants? Possibly. Researchers are working on pan-sarbecovirus or “universal” coronavirus vaccines that target conserved regions of the virus that mutate less.

Either way, this data demonstrates that SARS-CoV-2 wasn’t a typical bat Sarbecovirus that jumped straight to humans; the mutations would not have been tolerated in that environment.

SARS-CoV-2 had been practicing for a while.

I guess that means only one sacred cow is left standing - the furin cleavage site.

Let's see how the goal post will be moved once a sarbecovirus with a functional FCS is found in bats (or pangolins)... Give it time.

Cool preprint with promising results for a next-gen SARS-CoV-2 (potentially pan-sarbecovirus) vaccine, but my major takeaway more than anything here is how potent the selection pressure to escape antibodies is for SARS-CoV-2 variants.

11/ Simply acquiring a furin cleavage motif therefore does not automatically make sarbecoviruses better at entering human respiratory cells. Yet if sarbecovirus spikes frequently benefit from a furin motif, why is SARS-CoV-2 the only known sarbecovirus that has one?

Overall, the lineage had a whopping 6 reversions to the consensus Sarbecovirus sequence. I believe this reflects adapting back to a GI environment.

Speaking of gain of function, the Nebraska lineage appeared 3 more times in the last month.

The SARS-CoV-2 Spike RBD looks more like a bat Sarbecovirus (RaTG13) than the Nebraska lineage.

Think our immune systems would recognize it?
