Here’s the paper, I couldn’t really find details of AI used with quick read.

But can’t really figure out how predictive AI alone could work for that. Predictive AIs tend to be better for sales than science or serious engineering. Useful for cyber though.

Anyway, the point is that totally unexpected factors (like ORF3a) can have major effects on S1/S2 cleavage.

It may be, for example, that the effect of ORF3a or some other sarbecovirus protein partially explains why an FCS is not tolerated in the GI tract of bats, but generally favored in merbecos.

As of yet I know of no sarbecovirus other than SARS-CoV-2 that had it, but we recently discovered a furin cleavage site in a hibecovirus, the sister subgenus of sarbecovirus.

Linfa identified what was needed but overstated what it would prove. An FCS in another sarbecovirus would show natural acquisition in that subgenus. SARS-CoV-2 still aligns with the unusual research underway, lacks an identified animal host, and hints at contained evolution such work might produce.

I am sure the Coal Baron is magically gonna reverse his position on lab leak when a Sarbecovirus with a functional FCS at S1/S2 is found, right?

Of course not, he will double down and claim that when nature created it, isn´t it so much more likely the lab found it secretly and used it as template?

There are already plenty of bat BetaCoVs with FCS's. There's an alignment in the paper that shows several of them.

The "no FCS in sarbecoviruses" was never the slam dunk some presumed it to be. Now all or most of 3/5 BetaCoV bat subgenera have FCS's.

I don't think anyone seriously entertains the notion that WIV engineered >1000 nuc changes into the SARS-2 genome. But the only way around proposing this is to suppose that WIV secretly had a much more closely related sarbecovirus.

But in that case, why haven't we found it? Not to say...

Why would respiratory adaptation point to a natural origin?
DEFUSE proposed an FCS at S1/S2 in a sarbecovirus, then tests in human cells and humanized mice. Grow a virus in airway tissue and you get airway adaptation. That is not evidence against a lab.

If an FCS is deleterious in bat sarbecos, the natural route gets harder, not easier.

Then the first S1/S2 FCS in a sarbecovirus still appears in Wuhan, a year after they proposed putting one there.

That coincidence does not go away because the spike also needed longer NTD loops.
