Week 4 of HTGAA (Protein Design) is done & I'm amazed!

I was always intimidated by 3D protein models in papers & never thought I'd understand them one day!

Today, I loaded a PDB file into PyMOL, saw the alpha helices & beta sheets of my Tyrosinase enzyme.

This is so cool. 🚀

The enzyme responsible for his darker coloration is tyrosinase, which is one of the most important enzymes controlling the rate of production of melanin. When tyrosinase is hot, it binds tightly to an inhibitor, and can no longer perform its role in synthesis of melanin.

Plating a plant optimized tyrosinase plasmid onto LB with kanamycin 25mg/L, L-Tyrosine 500mg/L and Copper Sulfate 40mg/L. Let's see if it perks up melanin production in a day or two. Patched 4 colonies to start.

Tyrosinase expressing cells growing nicely at 30C! About 21 hours into growth. My experience back in 2020 was that it took a few days to really get melanin going. Fingers crossed for nice dark patches soon 🍫🦠

Das endoplasmatische Retikulum kam gerade endlich mal mit einer Funktion vor und nicht nur das gibt's und klingt witzig wie sonst immer!
Es synthetisiert zum Beispiel Tyrosinase.

Fortunately, most skin bleaching products don't involve caustic bases that turn skin cells into soap. They're things that exfoliate and/or block melanin or tyrosinase, as far as I know, and sometimes anti-inflammatories.

Working on that! The tyrosinase i have has a rubisco small subunit chloroplast tag, since tyr is mostly in cp. But if RUBY works then why not! Both run off Tyr. Vicia faba has a weird nitrogen-free alomelankn that looks black.

I suppose the petunia petals don’t have chloroplasts, but rather leucoplasts. Maybe the tyrosinase still gets in and does the job 🤔

FYI - we find it very hard / impossible to clone a tyrosinase in E.coli with a medium or strong constitutive promoter on high copy plasmid.
