MucD regulates alginate biosynthesis through the proteolytic control of AlgX and AlgK in Pseudomonas aeruginosa
by Yujun Jiang, Xin-Fu Yan, Rya Ero, Chao Wang, Kanaga Sabapathy, Yong-Gui Gao Pseudomonas aeruginosa is an opportunistic human pathogen capable of infecting a wide range of tissues and organs. Its persistence during chronic infection is strongly associated with biofilm formation, which depends on extracellular polysaccharides such as alginate. The HtrA-like periplasmic serine protease MucD is a key regulator of bacterial virulence, stress response, and alginate production, yet its molecular mechanism has remained largely unclear. Here, we discovered the alginate acetylation and export proteins AlgX and AlgK as MucD substrates, and characterized their degradation by mass spectrometry and bioinformatic analysis. We further determined the cryo-EM structure of MucD bound to an AlgK-derived substrate peptide, offering atomic insights into MucD oligomerization assembly, substrate recognition, and specificity. Together with structure-guided mutagenesis and biochemical assays, our results revealed that MucD proteolytic activity is governed by an equilibrium between a resting 12-mer and an active trimer. Crucially, we demonstrate that MucD represses alginate biosynthesis post-translationally, in addition to its previously implicated role in transcriptional regulation. These findings define a distinct activation mechanism and regulatory function for MucD and provide new insight into bacterial HtrA-like serine proteases.

Y'all, when you bring alginate to a job interview, be sure you don't mix it beforehand. Otherwise you're not going to make a good impression.
Folks, when you bring a laser to a job interview, do NOT bring one that has a poorly aligned crystal. It’s just not bright.
I’m sorry, but if you bring gypsum - or any damn mineral below 3 on the Mohs scale at all - to a job interview, you’re disqualified. It’s not hard, people.

Spherification looks like magic, but it's just sodium alginate meeting calcium chloride—a gelling reaction industrial food makers had used for decades. Adrià didn't invent the chemistry. He put it on a tasting menu and called it art.

Marie-Antonin Carême Wait—that's excluded too. Let me reconsider. Ferran Adrià — Spherification and the Alginate Bath
Ferran Adrià's use of sodium alginate and calcium chloride led to a breakthrough in liquid spherification, revolutionizing fine dining.

me at 20: I should buy this alginate casting kit and make a cast of my dick! just like Jimi Hendrix!

me in my 40s, looking at said unused kit: can I even hold an erection at this point?

Furthermore, the inclusion of fiducial fluorescent beads embedded within the alginate discs allowed us to find and track the same cells over time. (6/n)

Part of the novelty of this method is the embedding of protoplasts in alginate discs that fit into the wells of a 24-well plate. This format “enables high throughput experimental design: a single isolation yields more than 24 discs, each potentially representing an independent treatment.” (4/n)

Mix a diluted alginate solution (colored so you can watch the reaction) and drop it into a calcium lactate solution with a deep spoon; the alginate polymerizes instantly at the boundary between the two liquids, sealing a pocket of water inside.

@alexdainis.bsky.social shows how, using a bit of kitchen chemistry. Sodium alginate, a powder derived from seaweed, reacts with calcium lactate to form a gel wherever the two meet.
