As we age, our immune cells become less effective at clearing away senescent cells. A study published in Nature Aging suggests that a decline in chaperone-mediated autophagy impairs this cleanup process in macrophages, potentially contributing to age-related inflammation and disease.🧪
Decline of chaperone-mediated autophagy in aging impairs macrophage clearance of senescent cells - Nature Aging
This study looks at how the decline in chaperone-mediated autophagy in aging alters senescent cell properties leading to an impairment in their immune clearance. Utilizing proteomic and metabolomic analyses, the authors show that restoring autophagy enhances macrophage function and reduces senescent cell accumulation.

Restoring chaperone-mediated autophagy in macrophages is mechanistically stronger than merely correlating senescent-cell load with age. The next boundary is tissue and organism specificity: changing senescent-cell persistence may have context-dependent effects.
