Excited to share our new paper, now published in Nature Cell Biology! 🎉

We asked a fundamental question in cell biology: How do cells maintain polarity while migrating? Especially how do cancer cells navigate confined spaces?

Nature research paper: Fibroblastic reticular cells direct the initiation of T cell responses via CD44

Fibroblastic reticular cells direct the initiation of T cell responses via CD44 - Nature
CD44 expressed by fibroblastic reticular cells in secondary lymphoid organs regulates trafficking of dendritic cells, and thus has an essential role in the priming of T cells and the adaptive immune response.

This is interesting, when CD44 pulls in hyaluronan, it can also bring in bound iron, increasing intracellular iron, which may then reinforce CD44 expression.
CD44 regulates epigenetic plasticity by mediating iron endocytosis - Nature Chemistry
CD44 is a cell-surface adhesion receptor associated with many biological processes that rely on cellular plasticity. Now, CD44 has been shown to mediate endocytosis of iron-bound hyaluronates. Further...

A signaling axis promoting glycolysis in colorectal cancer cells, driving extracellular lactylation of CD44 & impairing T cell anti-tumor action🧪
Extracellular CD44 lactylation impairs CD8+ T cell function in KRAS-mutant colorectal cancer - Nature Metabolism
TRIP6 phosphorylation triggers a signalling cascade that promotes glycolysis in KRAS-mutant colorectal cancer cells. The resulting lactate accumulation drives extracellular lactylation of CD44 on CD8⁺ T cells, thereby compromising their anti-tumour activity.

A new #ScienceImmunology study demonstrates that T helper 17 cells upregulate CD44 and S1PR4 to transform into disease-promoting exTH17 cells in the synovium, which matches findings from human patients with rheumatoid arthritis. https://scim.ag/4nH90bi
TH17 cells converted into exTH17 cells sustain rheumatoid-like IL-17–independent inflammatory arthritis
ExTH17 cells drive IL-17–independent chronic SKG arthritis in mice through up-regulation of CD44 and S1PR4 and cross-talk with FLS.

CD44 is involved in cell adhesion, migration & signaling.
It is key in inflammation and immune diseases & cancer.

Good news: most commercial CD44 antibodies are highly specific and work well for:

• Western blot
• Immunofluorescence
• Flow cytometry

1/ An idea I'm kicking around:
The inhibitory neuron produces a large HMW-HA pericellular coat.
Astrocytes bind the HA coat through CD44 receptors.
The astrocyte mechanically probes the HA matrix through CD44.
