At this point it is important to note that other groups have described mtDNA release through endosomes (@dplamartin.bsky.social and @lauraenewmanphd.bsky.social in Gerry Shadel's lab at the time) as well as mtDNA release through MDVs (@prudentlab.bsky.social and
@mitodynamics.bsky.social).

In addition to the endosomal mtDNA release linked to myopathy. We also saw that some cell lines had smaller mtDNA puncta that were outside of the mitochondrial network but were not inside of endosomes.

It turns out these smaller mtDNA puncta were inside of mitochondrial derived vesicles (MDVs).

In the second paper (doi.org/10.64898/202...), we show that MFN2 variants that cause myopathy promote mtDNA release via endosomes, which leads to increased inflammation via TLR9 and cGAS-STING pathways. We believe this inflammation is what drives the myopathy in the patients.

Yes, if the big purple cavern is a nucleus then the cilia would be short, but plausible. PCD can do that. It would mean the scale bar’s too small though; mammalian nuclei in tissue are usually 10 um diameter, or smaller. The “nuclei” would likely be over-interpreted and wrong-colored endosomes.

WO2026178657A1 describes pH-responsive amphipathic peptides that become cationic in acidic endosomes, promoting cytosolic cargo release. In mRNA-LNPs, 1% FSP003D-CLS increased liver/spleen reporter output ~18×/~20×, with no obvious distribution shift. Escape and toxicity remain to be confirmed.
