Deletion of a distal IRF4 element prevents inflammation-induced reprogramming of human regulatory T cell fate
To investigate whether human Treg cells are susceptible to losing lineage identity, CD4+CD25+CD127lo/− fluorescence-activated cell sorting (FACS)-isolated primary human Treg cells (Extended Data Fig. 1a) were subjected to repeated cycles of in vitro CD3/CD28 stimulation in the presence or absence of proinflammatory cytokines (Fig. 1a). As a preliminary gauge of Treg cell identity, we focused on FOXP3 and HELIOS, a key pair of TFs with well-established, complementary roles in regulating Treg cell fitness and suppressive function13,14 (Extended Data Fig. 1b). As human conventional CD4+ T (Tconv) cells transiently express FOXP3 following T cell receptor (TCR) activation, expression profiles were evaluated 7 days into each stimulation cycle, after FOXP3 levels had mostly waned in non-Treg cells15. Previous studies on the impact of inflammatory cytokines on human Treg cell lineage stability have implicated a negative role of IL-1214,16, whereas the effects of IL-6 have been more controversial17,18,19,20. We thus started with unbiased comparisons of Th1-, Th2- and Th17-driving cytokines, singly and in combinations (Extended Data Fig. 1c). We found that the Th17-promoting combination of IL-6, IL-1β and IL-23 was the most consistent at inducing FOXP3 and HELIOS silencing (Extended Data Fig. 1c). This paper is a good example of the “if all you have is a hammer, everything looks like a nail” mindset. In an allograft setting,the authors observed that Tregs do not migrate to the graft unless Teff cells go there first, & they interpret this as something that needs to be fixed..

We need to look at the other plots as well. Here, Foxp3+ cells remain dominant, with no clear emergence of T-bet+ T cells. The Tregs are RORγt+ here (top plot), so a T-bet shift within the Treg population, as you suggested, seems unlikely.

7/ Even more interesting: these antigen-specific Tregs remained stable after DSS-induced inflammation or infection. Foxp3-negative cells emerged substantially only when IL-10 deficiency was combined with DSS.

5/ But is this really a new observation? Previous work already pointed in this direction. Mark Jenkins' lab found that gliadin-specific gut T cells were predominantly Foxp3+ Tregs, with essentially no conventional effector population.

Roinflammatory macrophage infiltration, enhanced FoxP3+ Treg recruitment and less profibrotic signaling are interesting, but "the AI told me" leaves zero traceability or audit trail, cant drive automated CAPAs or reviewability in teh QMS.

Just finished reading this. A very interesting review summarising a series of recent studies, largely from the Sun Hur team, on two immunity-related transcription-related proteins, FOXP3 and AIRE, both of which homo-multimerise in interesting ways.
