From lab reagent to metabolite: the riboswitch ligand guanidine as a relevant compound in bacterial physiology | Journal of Bacteriology
More than two decades ago, the first naturally occurring mRNA elements regulating gene expression by directly binding target ligands, termed riboswitches, were discovered in bacteria (1–4). Since then, more than 55 classes of riboswitches have been characterized (5, 6), advancing our general knowledge of gene regulation and stress response, but also unveiling unexpected biological roles for compounds whose relevance had remained elusive. A notable example is the discovery of riboswitches recognizing c-di-AMP and c-AMP-GMP (7, 8), which hinted at important biological functions for the two molecules that were later revealed to act as second messengers in key cellular processes such as biofilm formation, chemotaxis, sporulation, and virulence (9–11).

The origin of metabolic guanidine is largely a mystery. We suggest it is created when guanine-containing nucleotides are oxidised by molecular oxygen instead of being broken down into urea as purines....

Guanidine, known since 1876 for impacting muscle contraction, is linked to vertebrate RNAs in calcium signaling and neuromuscular functions. Discover more! PMID:40783389, Nat Commun 2025, @NatureComms https://doi.org/10.1038/s41467-025-62815-6 #Medsky #Pharmsky #RNA #ASHG #ESHG 🧪
Guanidine aptamers are present in vertebrate RNAs associated with calcium signaling and neuromuscular function | Nature Communications
Guanidine is a protein denaturant that is a widely used constituent in explosives, plastics, and resins. Its effects on muscle contraction were initially reported in 1876, which eventually led to the use of guanidine as a treatment for certain ataxia symptoms such as those caused by Lambert-Eaton disease. However, its mechanisms of therapeutic action remained unknown. Guanidine was recently found to be a widespread natural metabolite through the discovery of four bacterial riboswitch classes that selectively recognize this compound. Here, we report the discovery and biochemical validation of vertebrate members of guanidine-I and -II riboswitch aptamer classes that are associated with numerous genes relevant to neuromuscular function, mostly involved in Ca2+ transport or signaling. These findings suggest that guanidine is a widely used signaling molecule that serves as an additional layer of regulation of genes relevant to neuromuscular disorders. 150 years ago, guanidine was found to a

it looks to me like the word is derived from "guano"! "guanfacine" is the official chemical name and it's a guanidine derivative and guanidine was first derived from guanine which was first isolated from bird guano

also wait the G in DNA comes from bird poop????
