So how does HLH-30 prevent inappropriate developmental activity? Removing either BLMP-1 and CEBP-1, which become overactivated in the absence of HLH-30, partially suppresses the cell-cycle defects of hlh-30 mutants. This places HLH-30 upstream of a developmental brake during starvation. 7/8

Our results point to HLH-30, the C. elegans ortholog of TFEB. Without HLH-30, starved larvae inappropriately activate cell-cycle markers and seam cells can divide when they should remain arrested. In other words, HLH-30 is critical for enforcing quiescence during complete starvation. 4/8

How do cells prevent division when development has to stop during starvation? HLH-30/TFEB is a key factor maintaining cellular quiescence during complete starvation in C. elegans. 1/8

And HLH-30 does much more than control cell division. Loss of hlh-30 causes extensive transcriptional deregulation during starvation, particularly affecting genes involved in cell division and chromosome organization. 5/8

We also found a striking effect on nuclear organization. Starvation induces a characteristic reorganization of intestinal chromatin into two concentric rings. With the help of @daphne-cabianca.bsky.social we observed that without HLH-30, this architecture is disrupted. 6/8

Together, our work helps resolve a long-standing question:
What maintains cellular quiescence during complete starvation? Our results identify HLH-30/TFEB as a central part of that answer, linking nutrient deprivation to transcriptional regulation, chromatin organization and cell-cycle arrest. 8/8

Small medical update for those interested. Am in hospital at the moment getting some treatment and care. I don't usually talk about health stuff because A) it's boring, B) it doesn't really give anyone much to respond to other than 'hope you feel better soon' and C) it makes people feel bad. Sorry.

The Banff-registered fishing boat, Progress II, on the Rose Street Foundry's Thornbush Slipway at #Inverness. She was launched in 1907 but the slipway didn't open until 1908 so she's likely in for a refit or undergoing modification prior to being used as a patrol boat during #WWII
