I’m very excited to share my postdoc work in the @plaschkalab.bsky.social, where we investigated the very final act of splicing – spliceosome termination, revealing the stepwise dismantling of the spent spliceosome www.nature.com/articles/s41... (1/11)

BLM and FANCJ role in the response to G-quadruplex-dependent telomeric replicative stress - Communications Biology
Functional dissection of G-quadruplex–induced replication stress uncovers complementary telomeric roles of BLM and FANCJ helicases and identifies FANCJ loss as a vulnerability that increases telomere damage and sensitivity to G4-stabilizing agents.

To sum up, our work reveals how multiple RNA helicases and associated factors gradually unravel the RNA-protein network of the spliceosome after both successful and failed splicing events, sustaining accurate and efficient pre-mRNA processing. (10/11)

Using AlphaFold screens, we identified a set of novel termination factors, including 2 RNA helicases. We then purified the spliceosomes associated with these factors natively from human cells and determined structures of 2 new sequential termination intermediates: (4/11)

It turns out the intron-lariat spliceosome isn’t the end of the story! www.nature.com/articles/s41... We now resolve later terminating spliceosomes, showing how 3 RNA helicases and 7 dedicated factors gradually undo the intricate RNA-protein network of the spliceosome. (2/6)
Mechanism for the initiation of spliceosome disassembly - Nature
Cryo-electron microscopy structures of human and nematode spliceosomes show that the disassembly of this complex is initiated by the licensing of the disassembly helicase to act on the catal...

E. coli's DnaB helicase, key in DNA replication, efficiently unwinds DNA when correctly loaded, challenging the old view of limited ATP-motor coupling efficiency. PMID:42373613, Nat Commun 2026, @NatureComms https://doi.org/10.1038/s41467-026-73652-6 #Medsky #Pharmsky #RNA #ASHG #ESHG 🧪
Cellular replisomes are powered by flex-fuel motors for unwinding DNA | Nature Communications
DNA replication relies on hexameric, ring-shaped helicases to unwind parental DNA for supporting fork progression over tens of thousands of base pairs. Oddly, biochemical studies have suggested that, on their own, replicative helicases are rather limited motors that struggle to couple rapid movement to nucleotide turnover (typically believed to be solely ATP). Here, single-molecule studies reveal that when properly loaded, the Escherichia coli (E. coli) replicative helicase, DnaB, is a tremendously fast single-stranded DNA translocase that moves up to three times more rapidly than the replisome (3 knt/s). Translocation is highly processive, resistant to pulling force/high salt, and can displace short, 3’-tailed DNA duplexes without apparent changes in speed. Surprisingly, we find that the loader for DnaB, DnaC, can use any rNTP or dATP for depositing DnaB onto ssDNA and that the helicase itself also must hydrolyze nucleotide for stable loading. DnaB translocation also turns out to be s

In prokaryotes, Tiamat genes often neighbour helicases (~14% of Tiamats, ~33% of Tiamat-like), DNA methyltransferases, and nucleases. We speculate Tiamat nicks DNA, a helicase unwinds it and Tiamat degrades the ssDNA (untested). Some neighbourhoods may instead act on RNA. (8/11)

Tiamat is modular. Tiamat-like proteins often swap the nuclease for helicases, peptidases, or other effectors; some fungal ones carry Het-6, a non-self sensor that triggers cell death. On a GHL ATPase tree, Tiamat groups with human SACSIN, mutated in the ataxia ARSACS. (7/11)

Can yeast collected from a 250-year-old tree recreate colonial-style beer?

This question was explored by The Bochman Group, who study DNA helicases and genome integrity using the model organism Saccharomyces cerevisiae.

At the center of CPUG is MUT-15, which connects the RDE-8/NYN-1 nuclease module to the poly(UG) polymerase MUT-2. MUT-15 has the two RecA-like domains characteristic of DEAD-box helicases but lacks ATPase activity: it is a pseudohelicase!
