#IDRs
While screening for motifs, we discovered that locally scrambled IDRs were functional, and found conformational dynamics rather than linear sequence predicted function. We then used IDR dynamics as a design target and generated functional IDRs de novo.

The IRS IDRS system that holds all the data on taxpayers for the last 10 years written in cobalt I’ve added to my list and I still can’t replace it.

My group will scale this system and build AI models connecting IDR sequence, dynamics and function. We'll then write synthetic IDRs into genomes to understand how specificity and pleiotropy constrain IDR evolution. We think this approach will shed light on the dark matter of the proteome.

Folded protein design is now routine, but ~35% of the human proteome lacks stable 3D structure. Synthetic intrinsically disordered regions (IDRs) could be used for cell engineering. However, without structure, the target for biologically functional disordered protein design is not obvious.

mRNA 3′ UTRs chaperone intrinsically disordered regions to control protein activity
Highly conserved mRNA 3′ UTRs act as co-translational chaperones for intrinsically disordered regions (IDRs), preventing inter-domain misfolding and enabling biogenesis of fully active proteins.
