IFNg is an important discriminator for LC. But be careful, IFNg will be present during many infections and inflammation responses. i.e. IFNg does not mean LC.

It also does not mean IFNg is the cause of LC, not that IFNg causes the symptoms. But, it may and can enhance them.

This paper confirms the increased IFNg in LC patients. IFNg is secreted by CD8 and Th1 cells stimulated by monocytes and MHC (physiological), and actively secreted. Recovery from symptoms correlates with a reduction in IFNg production.

Note: CD69 expression indicates effector cells.

Changes in IFNg in patients with LC do not correlate with symptom severity.

This paper again shows that LC is an inflammatory disease, with many inflammatory mediators involved, such as the powerful cytokine IFNg. It is not known whether neutralising IFNg will provide symptom relief.

Patients can spontaneously recover from LC, and some can recover after vaccination. This correlates with reductions in IFNg.

This means there is less immune activity and less IFNg production, but still does not reveal the cause and consequence.

Now in #Science #Immunology: Philips, O'Shea, &al. made mice w human pathogenic #STAT1 activating mutation. Mice had #IFNg -dependent #autoimmunity, but their innatelike lymphocytes produced suboptimal IFNg during viral infection. Exogenous IFNg🚫virus-induced lethality, suggesting possible therapy!

What mechanisms drive checkpoint inhibitor-associated interstitial nephritis (ICI-AIN)?
In our preprint, we identify CD8 T cells as a central player, inducing a proinflammatory myeloid cell state by IFNg and structuring lymphoid aggregates by CXCL13.

Although IFNg may not directly be causing symptoms, it can, also indirectly contribute. The cause of more IFNg remains unknown and involves the established hypothesis of latent viral reawakening, viral debris/delay/failure to clear as well as re-awakening or induction of autoimmune activity.

Inverted CD4/CD8 ratio persisted during the whole time. But CD8-monocyte comolexes and IFNg + TNF production reduced.

1 patient: causal inference cannot be established. But interesting for follow ups and more investigation.

and genetic issues such as tight control of cytokines expression such as for IFNg itself, T cells or the antigen-presenting cells.

IFNg stimulation induces SLAMF7 expression on myeloid cells in rheumatoid arthritis. SLAMF7 engagement (with recombinant SLAMF7) then leads to a strong increase in a large number of proinflammatory mediators. SLAMF7+ are found in a number of inflammatory conditions.

A recurrent observation in #LongCovid patients is the increase in immune activity, including the production of IFNg from T cells. Much in opposite to any claims of "exhaustion", these cells express activation markers, such as CD69, PD-1.
