Now out in Science! Cyclin-dependent kinases (CDKs) are key regulators of the cell cycle. In @vcushing.bsky.social's magnum opus, we use #cryoEM to figure out how the CDK-activating kinase recognises CDKs to fully activate them - a key step in cell cycle control.

New preprint 🚨 We systematically measured 17 million phospho-specific dose-response curves (133 kinase inhibitors × 5 cell lines) to decrypt the kinases that shape the human phosphoproteome. We show that drug perturbation potency (not effect size) links kinases to substrates while controlling FDR.
Did you ever come across a phosphosite in your proteomics data for which nothing was known? - I bet so!

We have developed a new strategy termed "potency coherence analysis" that leverages the drug potency dimension in decryptM to decode the kinases that shape the human phosphoproteome.

But also - gulp - there's a whole regulatory layer involving phosphorylation of RNA polymerase too (the enzymes that reads and transcribes DNA) by kinase enzymes. And there are seemingly many more of these transcriptional kinases than we thought.

Agree completely. This looks like a variation on good ol' staurosporine, which hits half the kinases known to man. This compound is great news, but surely not because it's some sort of selective inhibitor.
Can't argue with clinical trial results.
But if this compound is acting selectively through GSK3-beta, I'll... be beyond surprised.

Phosphorylation depends both on the sequence and on spatial proximity in vivo between kinases and their substrates.

2 papers in @science.org this week uncover wheat tandem kinase function. Working in parallel, the authors of each paper show that two different tandem kinases converge to activate the same NLR!
