#NRAs
Just published in Science Advances:

Mutations don't have the same cancer effect in driving development of melanoma at every age.

Across 5,472 tumors, we find BRAF V600E loses selective advantage with age, while NRAS Q61 gains it.

Age reshapes selection on melanoma driver mutations
Aging is a major risk factor for melanoma. However, it is unclear whether aging fuels tumor evolution by altering mutation rates, changing the microenvironmentally induced selection on drivers, or bot...

NRAS Q61H is uncommon, yet its estimated strength of selection also tends to rise with age. The NRAS pattern extends beyond one particular codon-61 substitution.

I suspect aging changes the tissue environment (fibroblasts, matrix, immunity), favoring NRAS-driven signaling over BRAF V600E-driven signaling. "Pathways" are really networks. Both activate MAPK, but differ in downstream effects. Why aging shifts their relative fitness remains to be tested.

Stage matters, but mutations still guide treatment in advanced melanoma: BRAF-mutant tumors can receive BRAF/MEK inhibitors, NRAS-mutant tumors may soon be targetable with tailored RAS therapies. Our findings suggest age may further refine treatment choices. But it has yet to be tested!

Though its uncertainty is substantial, NRAS Q61K likewise has an increasing modeled selection trajectory.

♋ 🧪 www.science.org/doi/10.1126/...

Selective effects are best estimated variant by variant—not assumed to be universal to a gene. But a similar pattern strongly reinforces a conclusion.
November 7, 2026 at 1:04 AM
NRAS Q61L shows the opposite pattern: both prevalence and estimated selection increase with age.

A mutation strongly favored in a melanoma from an older patient need not be strongly favored in a melanoma from a younger patient.

Nice picture. I went there in 2012 with the Regional Furniture Society. I subsequently found the Apethorpe letters in the NRAS 217 catalogue at Register House, Edinburgh, and Lord Moray agreed for me to read them.

Repubs have been systematically building a totalitarian Regině for my entire life, at least since Nixon. Gengrich's Permanent Republican Majority. McConnell's court stuffing. The NRAs funding of law degrees.

It smart, strategic, and effective.

Huge congratulations to Helen for completing the Atlantic Coast Challenge in support of NRAS.

Reflecting Helen said: “My sister was diagnosed with RA at the age of 39 while raising young twins. Witnessing her courage and resilience in the face of such a relentless disease has been inspiring.”
