New EASD study highlights red meat sugar as independent risk factor for type 2 diabetes
Preliminary results of an observational study being presented at the Annual Meeting of The European Association for the Study of Diabetes (EASD) in Milan, Italy (Sept 28 – Oct 2), reveal that a sugar called dietary N-glycolylneuraminic acid (Neu5Gc), which is common in red meat, is independently associated with a 63% higher risk of developing type 2 diabetes (T2D) in those with the highest versus lowest consumption, even after taking into account other T2D risk factors such as level of physical activity, overall diet, smoking status and other health conditions. The association was weaker but remained significant after also accounting for body mass index (BMI). Our findings support chronic inflammation as a specific mechanism linking red meat consumption with T2D, partly beyond the amount of red meat a person eats and partly independent of body fat." Dr. Leopold Fezeu, Study Lead Author, Université Sorbonne Paris Nord The analysis found that the higher the levels of Neu5Gc, the greater the risk of developing T2D. As Dr Fezeu explained, " Although we do not know exactly what levels of Neu5Gc trigger an immune response, our data suggest that higher intakes of meat-derived Neu5Gc are associated with a progressively higher risk of T2D."...

If we merely "replicated" mammalian meat, it would still lack fiber and be high in saturated & trans fats. If we cut this out, we may as well get a Beyond burger today. The human Neu5Gc gene mutation also turns animal cells into cancerous pathogens for humans and only humans.

We compared LC–MS with DMB–LC–FLR because the latter is the established method for sialic acid quantitation.

The LC-FLR background limited NeuGc detection to 2 orders of magnitude, while LC–MS reached ≥3 and retained the intact N-glycan context, clearly defining the limitation of the FLR approach.

Calibration curves turn “detected” into assay-specific quantitative claims.

LLOQ varied by MS site and glycan. The widest dynamic range setup detected Neu5Gc-bearing N-glycans in neat human serum at ~0.01% of total sialylation.

This also explains the varied reporting of NeuGc in human samples

At low intensity, a precursor m/z alone is not enough.

We combined isotopic fit, PGC retention and MS2 to separate true Neu5Gc signal from noise, observing idotp decreasing with concentration.
Search rules should reflect the fragmentation method: re-CID does not retain the diagnostic 306.08 ion

To test low-abundance Neu5Gc in a realistic background, we serially diluted a Neu5Gc-rich N-glycome into human-serum N-glycans
The six-sample set, leveraging matrix-matched calibration curves (shout out @lindsaykpino.com) spans 4 orders of magnitude and ensures results are quantitatively informative
