In a new Comment in Nature Genetics, Caroline Hendry and Yong-hui Jiang of the Department of Genetics at Yale School of Medicine examine the FDA’s new draft plausible mechanism framework for individualized therapies targeting rare genetic diseases. Together with Jiangbing Zhou of Yale Neurosurgery and Biomedical Engineering, they assess both the promise of this framework and the groundwork needed to put it into practice. They argue that the guidance is best understood not as a green light, but as a detailed map of the work ahead—including building robust natural history data, validating biomarkers and assays, characterizing delivery platforms, defining patient-centered outcome measures, and developing scalable manufacturing infrastructure. Their perspective highlights the importance of early collaboration among researchers, regulators, clinicians, patient communities, foundations, and industry to realize the potential of individualized genetic medicines. In January, my employer apparently poked a hole in [protective layer guarding spine] during neurosurgery. In June, they fired me.

Noninvasive decoding of typed sentences from human brain activity | Nature Neuroscience
Modern neuroprostheses can now restore communication in patients who have lost the ability to speak or move. However, implanting these invasive devices comes with risks inherent to neurosurgery. Here we introduce a noninvasive method to decode the production of sentences from brain activity and demonstrate its efficacy in a cohort of 35 healthy volunteers. For this, we present Brain2Qwerty, a new deep learning architecture trained to decode sentences from either electro- or magnetoencephalography, while participants typed briefly memorized sentences on a QWERTY keyboard. With magnetoencephalography, Brain2Qwerty reaches, on average, a character error rate of 29% and substantially outperforms electroencephalography (character error rate: 65%). For the best participants, the model achieves a character error rate of 18%, and can perfectly decode a variety of sentences outside of the training set.
