Read length: I think the main inefficiency here might be the synthesis part, where if you start with a 5000bp molecule, only the first 500-1000bps are "expanded".
Key questions for me are:
1) Read length - how long can these actually get before they tangle, fracture or crash out of solution?
2) Raw error rate - how accurate can the novel polymerases get?
3) Is there any plausible pathway to detecting modified bases?
