Anti-viral drug obeldesivir has been shown to prevent against fatal ebola infection in rhesus macaques, which could greatly reduce the cost of managing ebola outbreaks and save lives 🧪

Very promising to see that Obeldesivir, an orally bioavailable nucleoside analog prodrug was effective against a lethal challenge of Marburg virus in a primate model of disease when administered within 24 hours of infection for a 10 day course 🧪

Oral dosing of the nucleoside analog obeldesivir is efficacious against RSV infection in African green monkeys | Nature Communications
Respiratory syncytial virus (RSV) is a significant cause of morbidity and mortality in high-risk populations. Although prophylactic options are available, there are no effective oral therapeutics for RSV infection. Obeldesivir (ODV) is an orally bioavailable prodrug of the nucleoside analog GS-441524, which is converted intracellularly to its active nucleoside triphosphate and inhibits the RSV RNA polymerase. Here we report the potent antiviral activity of ODV against geographically and temporally diverse RSV A and B clinical isolates (EC50: 0.20–0.66 μM). Resistance selection studies with ODV and GS-441524 against RSV identify a single amino acid substitution, I777L, in the L polymerase with reduced susceptibility (3.3-3.8-fold) to ODV and GS-441524, indicating a high barrier for resistance development. In an African green monkey RSV infection model, once-daily oral ODV doses of 30 or 90 mg/kg initiated ~24 hours post-infection significantly reduces log10 viral RNA copies/mL × day are

Post-exposure prophylaxis: Obeldesivir (oral antiviral) promising but needs a clinical trial first
None approved. All need ethics review, regulatory sign-off, import permits etc.

New research shows there's real promise that Gilead's antiviral #obeldesivir could be used to treat #Ebola Sudan & possibly other types of Ebola & #Marburg. It could make responses far easier: it's a pill, it doesn't require cold chain & it's not expensive to make. www.statnews.com/2024/03/14/e...

Antiviraltabletten Obeldesivir hade viss effekt i Fas 3-studier, men ingen kioskvältare vad jag kan förstå.
Obeldesivir är en prodrog (dvs kemiskt paketerad till ett förstadium i tablettform) av antiviralen GS-441524 som i sin tur är en metabolit av Remdesvir.

Texas researchers found that the oral drug obeldesivir (ODV) gave 100% protection to rhesus macaques and 80% to cynomolgus macaques against lethal Ebola when given 24 hours after exposure.

Obeldesivir, a SARS-CoV-2 antiviral, showed low risk of drug interactions in Phase I trials.

Minor exposure changes were not clinically relevant, and the drug was well tolerated, supporting its broad antiviral potential.
