I remember the first time I saw a canonical married gay couple in a PFS1 scenario. I did a double take, not because I was against it, but just because I was so used to it. They were the first tabletop company I saw to do that. To be clear, I support them in this.

CDK4/6 inhibitors (CDK4/6i) improve outcome in patients with advanced estrogen receptor-positive, HER2− breast cancer. The phase 3 SONIA trial compared the addition of CDK4/6i to first- versus second-line endocrine therapy for time to disease progression after second-line treatment (progression-free survival after two lines of treatment (PFS2)), as well as for secondary outcomes overall survival, PFS after one line of treatment (PFS1), health-related quality of life (HRQOL), toxicity and cost-effectiveness. No significant difference in PFS2 was observed; however, on an individual patient level this may be different. and figured out the whole pathway, particularly characterizing Pfs1, testing the vab-1/ephrin pathway, and very importantly, solving the mystery of maco-1’s role in PF15’s effects and epistasis, which made the whole thing finally make sense.

We were surprised to find that maco-1 gene expression is downregulated in PF15-trained worms, despite not being the direct match target of Pfs1 – suggesting that vab-1 is upstream of maco-1 expression.

So how does this relate to our previous findings about maco-1? As we found before with PA14 and P. vranovensis, maco-1 mutants/RNAi naively avoid PF15, and cannot learn further upon Pfs1 treatment.

Pfs1 treatment alone induces avoidance in P0-F4, and daf-7 is induced in the ASI neuron, as we had seen before.

Using genomics, logic, and screening, eventually we figured it out, and that was also weird: a small RNA (Pfs1) has a 16nt perfect match to vab-1, a gene that is best known for its developmental role in axon guidance via ephrin signaling.
