☕ @majarad.bsky.social et al. show that phosphatidylinositol 3-phosphate (PI3P), produced by the phosphatidylinositol 3-kinase PIK3C3, is formed on ER domains after lysosome damage. ER domain PI3P helps to recruit the ATPase DFCP1, which promotes lysosome repair.

Our new publication in Nature Cell Biology 🔬

How cells reseal damaged lysosomes: Ca²⁺ leak → VPS34 makes PI3P on the ER → recruits DFCP1 → clusters VPS13C lipid-transfer bridges at ER–lysosome contacts to patch the membrane. No DFCP1 = failed repair.

Our study on DFCP1 and lysosome membrane repair is live! We uncover a previously unrecognized role for omegasomes in the cellular response to lysosomal membrane damage, revealing another layer of regulation in how damaged lysosomes are repaired.

⭐ Our data suggest that targeting PIK3C3 activity relocalize the lysosomes from the cell periphery to the perinuclear region leading to lower mTORC1 activity in BC cells.

⭐ We revealed that dormancy-prone or dormant breast cancer (BC) cells might be eliminated by targeting PIK3C3 (the 3rd class PI3K), hence less #metastasis burden in #breastcancer.
