One of the most exciting applications of #miBrains in the Blanchard Lab is the prospect of personalized medicine.

Because miBrains start from human cells, they can be engineered to match an individual donor's genetic profile – starting from something as simple as a skin biopsy.

not only imaging pericytes/endothelial connectome at EM level, but we have high interest in capturing and quantifying pericyte mitochondria as well. this would be super fun to do this with you and @choroidlab.bsky.social
how difficult would it be to see/send a small snippet of EM volume ?

Emergence of fibrotic pericytes and their transcriptional regulation in pulmonary fibrosis | Nature Communications
Multiple cell types have been implicated in pathogenesis of pulmonary fibrosis with pericytes emerging as a new focus due to their role in promoting fibrotic remodeling. Fibrotic environment changes normal pericyte functions, but transcriptional programs regulating pericyte transition towards fibrotic state remain unclear. Utilizing single cell RNA-sequencing of human pulmonary fibrotic lungs and mouse genetic models, we identified a unique cluster of fibrosis-associated pericytes which was not present in normal lungs and exhibited a distinctive transcriptional signature indicating transition of normal pericytes to a fibrotic state. FOXF1 was identified as one of the transcription factors decreased in fibrotic pericytes. Pericyte-specific deletion of Foxf1 increased severity of pulmonary fibrosis in bleomycin mouse model as demonstrated by reduced survival, impaired lung functions, increased body weight loss and increased fibrotic remodeling. Pericyte-specific overexpression of Foxf1 a

🌱 APOE4 may do more than raise Alzheimer's risk. A Cell study links it to a vascular shift: pericytes become myofibroblast-like cells, secrete fibronectin and promote vascular amyloid. Blocking TGF-beta reversed this in models. www.cell.com/cell/fulltex...

A pericyte-to-myofibroblast transition links APOE4 to cerebrovascular degeneration
APOE4 causes a pericyte-to-myofibroblast transition that promotes cerebrovascular fibrosis and perivascular amyloid accumulation. TGF-β inhibition reverses this pathological remodeling and thus presen...

the apoe4 gene, a major alzheimer's risk, damages brain blood vessels by turning pericytes (vessel-stabilizing cells) into scar-formers, leading to amyloid buildup. blocking a protein called tgf-beta protected these cells!
