Pfft, this is easy! you just need to encode the Epstein list using peptides in a QconCAT like protein . Express and analyze using LCMS and then put the results in your jupyter QC notebooks. Easy peasy . Oops was I not supposed to say that?

We combined this and the QConCat concept to create a plasmid that was cut open with a 3 base pair overlap using AdeI.

We encoded the peptide sequence of interest in DNA with a 3 base pair overlap on each end that matched the overhang in the plasmid, typically a lysine.

Myself and Hamid Mirzaei experimented with QConCat constructs but we struggled with the differences in response from each peptide that pushed the dynamic range of the MS systems at the time.

Question for #teammassspec here. Are internal standard peptides like AQUA or QConCat still in use commonly? I haven’t worked in this space for a number of years but worked on a project that never saw the light of day that I may just open source the concept here.
