Transgenerational epigenetic inheritance without RdRP...!

Lack of RdRP was one reason people argued epigenetic phenomena in nematodes (which do have RdRP) are fascinating but maybe not relevant to other animals (which don't have RdRP). But now....
In this new work we show that planarians establish long-lasting and heritable small RNA–based gene regulatory states despite lacking canonical RNA-dependent RNA polymerases and nuclear RNAi machinery.

PB2 is one of 3 subunits (with PA and PB1) that make up the viral RNA-dependent RNA polymerase (RdRp). The RdRp both makes mRNA that’s translated into viral proteins and copies the viral genome segments in the process of virus replication to package into progeny virus particles.

There has been enough opportunity for this substitution to occur, though (same for rdrp adaptation btw). I suspect that there is some other biological precondition we only poorly understand.

Contrary to what many people think we know very little of why one virus turns pandemic while others do not.
After 8 months of peer review, our manuscript was published today @nature.com showing that bovine H5N1 viruses bind poorly to human-type sialic acid receptors. We now know that the virus is only 1 HA substitution away from efficiently binding human receptors.

(1) we need to see what the german intelligence agency has, but the inferrence is that they may have a copy of Ping Yu's masters thesis documenting diverse partial RdRp sequences from bat samples of diverse SARSr sarbecoviruses.
Got it, thanks. If this what they are basing their intelligence on then they are not very smart. A paper describing this work was submitting in 2018.

What’s more is that glycoprotein usage is not congruent with our Nidovirales polymerase (RdRp) phylogeny, suggesting a complex history of glycoprotein switching throughout this order.

So what do these mutations do? Well, because they are in the PB2 subunit of the RdRp, they change how the RdRp works.

D701N increases polymerase (RNA copying) activity & enhances viral replication, as shown by virology legend Hans-Dieter Klenk et al.

Hey! That's me! Check out my latest where I develop a method for pinpointing where mutation spectra change along phylogenies. Striking, nodes with large changes are significantly enriched for protein changes in the RdRp (catalytic domain of the polymerase) suggesting a causal role.
