I'm pleased to share our most recent study led by Matija Zelic. We profiled human ALS spinal cords to uncover neuroinflammatory microglia and astrocyte states and show a key role for Ripk1 as a contributor to the pathological tissue environment in ALS.

Repression of RIPK1 kinase by INPP5D inhibits expression of diverse proinflammatory mediators and late-onset Alzheimer’s disease risk factors
The function of the Alzheimer’s disease (AD) risk gene INPP5D in microglia remains unclear. Xie et al. identify INPP5D as a direct regulator of RIPK1 kinase, known to be implicated in mediating neuroinflammation. They show that INPP5D deficiency in aging activates RIPK1-dependent diverse innate immune signaling, promoting TDP-43 pathology and neuronal loss associated with AD and motoneuron degeneration, such as amyotrophic lateral sclerosis (ALS).

🧠 Low brain glucose may be more than a symptom of Tau disease. Hypometabolism and p-Tau synergize to trigger neuronal necroptosis via a p-Tau-RIPK1 hub. Restoring A20 or blocking the interaction eased brain atrophy in Tau mice. Preclinical, but striking.
