!!! Just out in @jacs.acspublications.org !
We made flexible, saddle-shaped COTs via double SPAAC.
They stack perfectly (SASA), bend elastically thanks to adaptive curvature + alkoxy chains,
and guide light through cm-long crystals.
Final PhD project — and my favorite.

Click To Stack: Shape-Assisted Self-Assembly of Unflattened Macrocycles into Waveguiding Elastic Crystals
Designing molecular crystals for practical applications requires precise control over intermolecular interactions. We report a click-to-stack strategy to enable facile construction and systematic structural modification of tetraarene-fused cyclooctatetraenes. These saddle-shaped molecules self-assemble into one-dimensional columns with exceptional precision, driven solely by weak van der Waals interactions. Single-crystal X-ray crystallographic and computational studies revealed fully eclipsed π–π stacking, reinforced by the shape complementarity of the negatively curved molecular contour. With alkoxy groups tethered, the antiparallel alignment of coaxial cable-like assemblies produces centimeter-long voidless crystals that exhibit remarkable structural durability under mechanical stress. These glow stick-like elastic crystalline rods display optical waveguiding properties, demonstrating the practical utility of highly anisotropic molecular assemblies with structural uniformity and resilience.

wow this is a really nice paper and a clever approach! the narrative carried me right along as my questions arose. congratulations. some unsolicited review: are picolyl azide substrates available? I love these for the kinetics and low copper requirements, and I find SPAAC reactions often need..

Thanks for the kind words, @fewill26.bsky.social @chemkritzer.bsky.social @rche54.bsky.social! I tend to focus more on reaction development & mechanism from Bertozzi (Staudinger), Bertozzi & others (SPAAC), Sharpless, Finn, Meldal (CuAAC), and Fox, Weissleder, & others (IEDDA) 1/2

Thank you so much, Carolyn! This would not have been possible without metabolic labeling and spaac…!:)

Strain-promoted azide–alkyne cycloaddition (SPAAC) chemistry is used to covalently conjugate dibenzocyclooctyne (DBCO)-labeled antibodies or antibody fragments to LNPs containing azide.
March 10, 2026 at 5:59 AM
The approach was validated on purified proteins as well as in living cells, including selective labelling of modified trastuzumab, concanavalin A, and cellular organelles.

Beyond traditional strain-promoted azide–alkyne cycloadditions by achieving orthogonality and rapid kinetics with fluoroalkyl azides - Communications Chemistry
Strain-promoted azide–alkyne cycloaddition (SPAAC) is crucial for bioorthogonal chemistry but suffers from slow kinetics and limited selectivity, complicating multi-target labeling. Here, the authors ...

🫆 #BioorthogonalChemistry enables labeling molecules in biological systems without disrupting their processes.

Among its key tools is SPAAC, a “click” reaction used in bioconjugation and live-cell imaging—yet its slow kinetics and limited selectivity complicate labelling of multiple targets.

Conjugating commercial click-modified DNA oligos also worked at the first shot without optimization, although at relatively low conjugation yields. We’re not so experienced with the chemistry, but apparently Cu catalysis isn’t great for DNA either, so probably switching to SPAAC would work better.

Small molecules were readily conjugated to all our example proteins: we show attachment of fluorophores & biotin, and establish that both classic copper-catalyzed, as well as @carolynbertozzi.bskyverified.social's copper-free SPAAC reaction are compatible with ADD tags & result in full labeling.

SPOCQ has the great advantage over SPAAC as the proteinogenic amino acid tyrosine offers direct access to a rapid and clean method for protein modification, especially to convert therapeutic antibodies into antibody-drug conjugates (ADCs) and bispecific T cell engagers (BiTEs) (vide infra).

With deltaH++ of <2 kcal/mol, these cycloadditions are entropy-driven. This has been mentioned before in the literature, but now we have the exact numbers, obtained by stopped-flow spectroscopic analysis!

Reaction rates in SPAAC but not in SPOCQ correlated to sp-sp3 bond angles!

We used a stable quinone and a range of strained cyclic alkenes and alkynes such as BCN and DBCO for in-depth kinetic studies of the SPOCQ click reaction (which is typically >100 times faster than the more commonly used SPAAC).
