Allosteric inhibition of human mitochondrial ClpP by hijacking the dordaviprone (ONC201) binding site - Nature Communications
ClpP is a mitochondrial protease and a validated cancer target. Here, the authors develop quinazolinone compounds that bind to the same site as the FDA-approved activator ClpP dordaviprone, yet they inhibit ClpP, reducing tumor burden in leukemia mouse models.

Our new preprint describes an allosteric glue. Here, "glue" refers to a compound that causes two proteins to stick together. Usually the glue touches both proteins but here it only has to touch one!

Happy to join bluesky.
My lab uses structural biology to investigate GPCRs. Specifically, we are interested in chemokine receptors and 1) how differences in ligand interactions leads to pathway-selective signaling, as well as 2) allosteric modulation via the intracelluar allosteric binding site.
