I think there is a need for a complete re-think for organoid modeling.

As long as the architecture of organoids that you produce is not directly compared to a 3D map of the organ (say, a fallopian tube) or disease to be modeled, it will be approximate.

We use CODA to map human endometrial tissue at single-cell resolution, a map that then serves as a blueprint to design a multi-compartment multicellular assembloid of the endometrium.

This spring, we reported in Nature a therapeutic approach for Timothy syndrome

This has been a long journey starting with making hiPS cell-derived neurons in 2D cultured 15 years ago when I was a postdoc & continued with creating in the lab #organoid and then #assembloid models of this disease

A clever forebrain assembloid model! Congratulations!
Ryan’s paper out in Neuron (bit.ly/3IICGFX). He derived bona fide parvalbumin (PV+) cortical interneurons from hPSCs, one the slowest developing neurons, and could capturing interneuron-related disease phenotypes linked to schizophrenia - including impaired gamma-oscillations - in his assembloids.

We will post soon two major papers on women’s health.

The first paper could change clinical practice for the early detection of ovarian cancer.

The second paper shows the creation of new cell assembloid that reflects the complex composition and architecture of endometrial tissue mapped in 3D.

In our new paper, we use a new 3D single-cell reference map to guide the design and validation of a new multi-compartment assembloid that mimics the function and architecture of the human endometrial tissue, exhibiting stromal decidualization.

Fake kidney. This lab-grown human kidney ‘assembloid’ is a step towards creating artificial kidneys. Tiny filtering units have formed around a central drainage tube, similar to the organization of a real kidney.

A 3D multi-compartment assembloid to study
combined immune cell infiltration and cytotoxicity

Contains different compartments with different ECM.

Especially useful to test infiltration and cytotoxicity of CAR T cell therapies.
