A guide to CAR T cell therapies: development, current status and future prospects | Nature Reviews Immunology
Since the first clinical approval in 2017, chimeric antigen receptor (CAR) T cell therapy has emerged as one of the most powerful modalities for redirecting the immune response against cancer. Building on decades of foundational discoveries in T cell biology and synthetic immunoengineering, CAR T cell therapy has transformed the treatment of B cell malignancies, resulting in durable remissions in patients with B cell leukaemias, lymphomas and multiple myeloma. Next-generation CAR designs are now expanding the reach of this approach into autoimmune disease and solid tumours. Innovations in gene editing, allogeneic manufacturing and in vivo delivery are improving the scalability, safety and accessibility of CAR T cell therapies, although challenges persist in overcoming antigen heterogeneity and tumour microenvironmental barriers and in promoting the long-term persistence of CAR T cells. In this Review, we summarize the key discoveries that laid the foundations for CAR T cell therapies a

New preprint with Zernicka-Goetz lab (@zernickagoetz.bsky.social) on lineage reconstruction in mouse embryos and stem cell-derived embryo models, led by Yunzheng Liu! (1/13) www.biorxiv.org/content/10.6...
Lineage tracing reveals asynchronous fate restriction in mouse embryos and stem cell-derived embryo models
How a flexible mammalian embryo gives rise to a reproducible body plan remains unclear, partly because cell lineages cannot be followed continuously in vivo. We combined DNA Typewriter lineage recording with tetraploid complementation to reconstruct high-resolution single-cell genealogies across mouse development and built FateVec, an analysis framework that infers fate-bias dynamics and lineage-associated transcriptional programs from reconstructed phylogenies. Loss of developmental potential proved progressive and asynchronous, with multiple lineages acquiring detectable fate bias before morphological diversification at gastrulation. Within the neural crest, most cells remained multipotent over an extended window, while independent founder clades differed markedly in restriction timing. Extending lineage recording to a stem-cell-derived embryo model revealed that cells with shared transcriptional identities can arise from distinct lineage relationships. We further found embryonic-lineage contributions to extraembryonic-like states, exposing an unexplored lineage plasticity. This lineage-resolved view of embryogenesis reveals that cell identity alone predicts neither developmental ancestry nor the timing of fate restriction. ### Competing Interest Statement The University of Washington has filed patent applications related to molecular recording technologies, in which J.C. is listed as an inventor. The authors declare that they have no other competing interests.

DNA recording system reveals how a single cell becomes a body
The recording covered the first 13.5 days of embryonic development, providing an unusually detailed look at how a complex mammalian body takes shape. (Full gestation in mice is 19 to 21 days.) The findings, published on October 8, 2026, in the journal Science, could help scientists better understand normal development and what happens when that process goes wrong. "We are interested in tracking how cells' early ancestry in an embryo influences their later fates. Exploring cell lineages helps decode normal development. Such research could eventually advance knowledge about congenital malformations, neurodevelopmental conditions, genetic disorders or cancer," said Dr. Jay Shendure, a professor of genome sciences at the University of Washington School of Medicine in Seattle and a Howard Hughes Medical Institute Investigator. Shendure led the research alongside Dr. Chengxiang Qiu, a molecular and systems biologist at Dartmouth College in New Hampshire. Both served as senior authors of the study. A DNA Typewriter Records the History of Living Cells The achievement was made possible by a technology called DNA Typewriter, developed by Shendure and Junhong Choi, who is now at Sloan Kettering Cancer Center. Rather than simply observing cells as they divide, the system allows scientists to preserve a record of...
