Second round of culturing took longer bc it's free of any pre-made aminos, but now even more evidence to support this sucrose based selection system works. Both media is M9 with 20mM Sucrose as sole carbon. cscA is expressing invertase, cscB flask expressing sucrose permease.🍬🦠

Plan for next week is to construct this vector, expressing the cscA sucrose hydrolase and cscB sucrose permease using a strong and weak promoter. Linking the two transcription units (TU's) into one module via golden braid. Goal is to see if permease is needed and measure growth.

Using my bacterial turbidometers, I'll grow various strains of lab E. coli with just the cscA, just cscB, cscA+cscB, and a neg control with identical plasmid sans sugar genes. I did a manual end point experiment with just cscA overexpression and saw strong growth only on cscA+.

And my favorite bit of evidence! The hydrolysis of sucrose yields glucose and fructose. The aerobic metabolism of glucose acidifies the media, leading to a color change in universal pH indicator toward red. The cscA flask is red while the cscB flask remained the default green.

The Cypher System Character Builder Is Now Live! - Monte Cook Games
Cypher System Character Builder opens to the public on Monday, the 8th of April. It will be free to use, and is a part of the growing Cypher Tools suite of online game tools. Using the Character Builder, players can generate Cypher System characters for any genre, drawing on character options from the Cypher System Rulebook and a full range of major Cypher System supplements. The entire line of Cypher System genre supplements is included, and content from new titles will be introduced as those titles are released.
