Chromatin organizes nuclear DNA, but whether it actively regulates whole-cell mechanics remains unknown. Here, we used NETosis to show that chromatin decompaction within the nucleus increases plasma membrane tension and cell volume, establishing chromatin as a regulator of cell mechanics.

We combined single-nucleosome imaging and 3D-SIM to reveal:
🔹 Euchromatin forms condensed domains, not open fibers
🔹Cohesin loss increases nucleosome mobility without decompaction
🔹Cohesin prevents neighboring domain mixing

We look forward to hearing feedback from the community on our #preprint where we used #NETosis as a platform to show that chromatin decompaction inside the nucleus directly regulates cell mechanics. This work identifies long-range biophysical impacts of chromatin in mammalian cells.

So, is it compaction or decompaction? Here is a notable alternative hypothesis: "Our data suggest that the process of..loop extrusion per se is not required for enhancer function..Rather, it is the chromatin compaction brought about by loop extrusion that may be the important factor to consider."
Plenty of work required to rigorously test the proposed hypothesis but my initial thoughts are this perspective by Adrian Bird in @cp-molcell.bsky.social may well be the most significant conceptual advance in the 3D genome organisation field in years:

3/Chromatin is compacted in apoptosis, might this sequester fragments? Histone deacetylation, which drives compaction in living cells, has been observed in apoptosis, but a role in apoptotic compaction was unknown. We inhibited apoptotic deacetylation which resulted in drastic chromatin decompaction

We measured single molecule ssDNA binding by SSB proteins from different systems, E. coli SSB, T4 gene 32, and LINE-1 ORF1p. All showed surprising initial compaction then decompaction. This fits well with your data: the increased inter-probe distances could reflect deformation/elongation of a TAD rather than uniform decompaction. Hi-C data (Fig. 8a) show that short-distance contacts decrease and long-distance contacts increase after cohesin loss. Thanks for pointing this out! 2/2
