This means that when cargo concentrations on an Endosome are high, then PI(3)P is also high and vice versa! And we could directly demonstrate this connection by live cell microscopy of single endosomes trafficking transferrin!

Endosome motility controls light-responsive reproductive development and secondary metabolite production in Aspergillus CurrentBiology
Endosome motility controls light-responsive reproductive development and secondary metabolite production in Aspergillus
Kumar, Allen, Oster et al. show that motility of PxdA-marked early endosomes in Aspergillus nidulans is required for light-responsive development and production of secondary metabolites. In the pathogen A. fumigatus, PxdA loss also alters secondary metabolites. This links organelle dynamics to reproductive fate and secondary metabolite output in filamentous fungi.

A study in Cell reveals the role for Prf1 in directly inducing ferroptosis in tumor cells.
🔹Perforin-mediated pore formation promotes transferrin receptor endocytosis.
🔹Endosome–mitochondria contacts facilitate mitochondrial iron accumulation.
🔹Higher Prf1 level is associated with enhanced survival.

First we need to establish how endosomal PI(3)P is organised. Using recombinant biosensors we can detect free PI(3)P and find that it is segregated into specific domains: High free PI(3)P on the bulk endosome and low PI(3)P in the retrieval domains, marked by Retromer component VPS35:

While the endosome release pathway clearly correlates with myopathy, we still don't now the relevance of the MDV release pathway to MFN2 disease - though it does lead to a 2-3 fold increase in cGAS-STING inflammation.
