Using cryo-EM and biochemical approaches, we characterise a novel INO80 binding mode (“Off-state”) on a -competent state when oriented toward the gene body, but an inactive Off-state when oriented toward the promoter.

Together, our findings show how DNA sequence can influence both INO80 binding mode and remodelling directionality, providing a mechanism for sequence-driven +1 nucleosome positioning.
