I had a tricky clone run that had a fusion protein containing kanamycin resistance while on an Ampicillin plasmid, that I needed to clone into a kanamycin plasmid. Did a patch plate with few colonies. Growth on kan (left) but not on amp (R) while still glowy is correct clone. Got em! PCR and seq now

Even with my love of omics, we've patched hundreds cells for an ongoing patch-seq project in Oxford and there just isn't a good alternative to an expert at a rig

Realized it would be fun to patch a Boundary Layer function with a seq where the seq controls the Bound input of the function (which opens lpg)

Seq out mult'd to a quantizer & pitch input so the higher the sequencer step the higher the pitch but the shorter and less loud the played note

Interesting. We observed similar shift of inter-trial activity between air puffs in a previous study: www.nature.com/articles/s41.... No humans and no Ketamine in there, only mice. But we can trigger the same shift with subcortical stimulation (hypothalamus/habenula projection).

Spindle neurons in human cortex possess distinctive firing properties and transcriptomic signatures | Nature Communications
Spindle neurons (SPNs), also known as von Economo neurons (VENs), are predominantly found in large-brained species including humans. SPNs in human neocortex are implicated in cognitive functions and affected in various brain disorders. Despite their unique morphology, whether SPNs possess distinctive electrophysiological properties and transcriptomic identities remains unclear. Here, we performed whole-cell patch clamp recording (134 cells) together with RNA-sequencing (patch-seq, 124 cells) on visually identified SPNs and pyramidal cells (PCs) in human cortical slices. Compared with putative extratelencephalic-projecting PCs, SPNs possess less complex proximal apical dendrites and emanate axons from the basal dendritic trunk. Additionally, they exhibit heightened excitability and characteristic firing properties, including robust bursting and persistent firing pattern. Patch-seq analysis revealed transcriptomic heterogeneity within SPNs, and identified their fingerprint genes. Collect
