Vermutlich ist es ein "Klassen-Effekt" der mRNA-LNP Plattform. Diese führt zu einer Aktivierung des angeborenen Immunsystems, was sekundär CD8 T Zellantworten verstärkt. Der beobachtete Effekt war in Kombination mit einer Checkpunkt-Blockade (PDL1-Antikörper) bei NSCLC und Melanom.

Biomarker in BrCa session has me feeling biomarker angst. Lovely data re quality of immune response to anti-HER2 mAb predicting outcome (Sotiriou). But how to reconcile concept w HER2-low ADC data? Also PDL1 doesn't predict anti PDL1 benefit in TNBC. 'Obvious' targets = poor predictors. #AACR25

Press release: phase III SKYSCRAPER-01 study negative for OS at final analysis. Adding anti-TIGIT tiragolumab to first line atezolizumab in advanced PDL1 high NSCLC did not improve PFS or OS. Now to dissect what happened between phase II and III.

PDL1 is generally very weak and heterogeneous, making it difficult to image (given its importance clinically). Huang et al. optimized a high-dynamic-range (HDR) imaging method for immunofluorescence to assess PD-L1 expression. 🧪

#ScienceSaturday CD28 is an important costimulatory molecule for T cells. However, in a recent study, Yang et al utilized a whole genome CRISPR screen in a breast cancer model to identify that tumors can also use CD28 signaling to inhibit T cells through upregulation of PDL1 on the tumor surface.
