Prognostic influence of small leucine-rich proteoglycans on serous ovarian cancer - Journal of Cancer Research and Clinical Oncology
Purpose Ovarian cancer is one of the most lethal cancers in women worldwide. To be able to offer successful treatment and improve the prognosis, knowledge of factors influencing the tumor microenvironment is indispensable. In this context, the influence of the extracellular matrix on tumor progression is increasingly recognized. Of note, preclinical data in cell line and animal models have suggested that several members of the small leucine-rich proteoglycan (SLRP) family are mechanistically involved in the regulation of tumor progression. We hypothesized that dysregulation of SLRP expression may have a prognostic value in ovarian cancer. Methods To distinguish whether this expression is altered in the cells themselves or in the extracellular matrix, quantitative Real-Time PCR was performed on ovarian cancer cell lines and complemented by analysis of CCLE datasets. We used Kaplan–Meier survival curves to investigate whether a high or low mRNA expression influences the survival of

GPC6 facilitates progression of SHH-subgroup medulloblastoma by enhancing Hedgehog secretion and signaling responses
<p>Medulloblastoma (MB) is the most common malignant tumor of the cerebellum in children. The Sonic Hedgehog (SHH) subgroup of MB (SHH-MB) is driven by aberrant activation of the SHH pathway; however, mutations in genes related to this pathway are relatively rare, posing challenges for therapeutic development. Glypican-6 (GPC6), a heparan sulfate proteoglycan, is highly expressed in SHH-MB. In this study, we demonstrate the synchronous expression of GPC6 with GLI family zinc finger 1 (GLI1) in both the developing cerebellum and medulloblastoma. GPC6 promotes cell proliferation, migration, and invasion in SHH-MB cell lines (DAOY and ONS-76). Consistently, GPC6 enhances the SHH pathway activity by upregulating GLI1 expression, supports ciliogenesis essential for signal transduction, and facilitates SHH ligand expression via extracellular vesicles. These findings suggest that GPC6 acts as a key regulator of SHH signaling and represents a potential therap

Increased transvascular retention of atherogenic lipoproteins in type 2 diabetes relates to their enhanced proteoglycan binding
Subendothelial retention of cholesterol-rich apolipoprotein-B–containing lipoproteins drives atherosclerotic arterial disease. In peripheral interstitial fluid from patients with type 2 diabetes (T2D), levels of such particles have been shown to be paradoxically reduced relative to those in serum, presumably reflecting their increased retention within the arterial wall. To identify possible mechanisms involved in lipoprotein retention in T2D, we obtained serum and skin blister fluid from such patients and matched controls, together with skin biopsies in a subset of individuals. In T2D, smaller LDL and VLDL remnant particles were more prominent in serum but not in interstitial fluid, reflecting their enhanced vascular entrapment. The interstitial-fluid-to-serum ratio of apolipoprotein-B was 58% lower in T2D than in controls (0.14 versus 0.33), concomitant with increased susceptibility for LDL binding to proteoglycans. The most marked differences were seen in patients with clinically evident cardiovascular disease. The degree of transvascular retention was positively related to the propensity of isolated serum LDL to bind aortic proteoglycans, both in T2D and in controls. Skin unesterified cholesterol levels were higher in patients with T2D relative to healthy controls. With aging, both proteoglycan binding and apparent vascular retention of LDL increased in controls but not in T2D, indicating that these mechanisms may also be relevant for atherogenesis in nondiabetic individuals.
