This is what happens when you remove the wall from plant cells. They become perfectly spherical and are called protoplasts.
These protoplasts were isolated from Arabidopsis and transformed with a vector to express cytosolic GFP (in orange). The chloroplast autofluorescence is in grey.

The protrusions of the chloroplast envelope are called stromules.

I wondered that as well. Usually these older protocols were root(callus)-derived protoplasts. I imagine that it's done to stress them less. For leaf protoplasts I never noticed an appreciable difference between dark/light

We’re back at the @dsw-isp.bsky.social annual meeting this afternoon where @emilieknight.bsky.social is presenting a promising protoplast-based method to decipher gene networks in wheat. Protoplasts are living plant, bacterial, or fungal cells that have had their rigid outer cell walls removed.

The death of protoplasts. I was told that I made a restaurant for fungi by keeping my leaf protoplasts in not fully sterile conditions. However, they still look pretty.

linet is a protoplast , colloquially referred to as an elf, but a human being all the same. i'm working on designs/lore for protoplasts and hope to post smth about them soon... linet himself has gone through a few design revisions and i'm still adjusting, along with the other charas designs

Another nice thing about ExPOSE is its versatility. We used it to observe biomolecular condensates in Arabidopsis protoplasts and transcription factors in maize protoplasts at an enhanced resolution. 🧵

Great work by bachelor student Martijn Posthumus during his 10-week internship. He built tagged versions of GFP that are transported to different cellular compartments. We will use it to study protein sub-cellular localization in plants.
