How does human AGO2 distinguish cleavable from non-cleavable RNA targets?

Our cryo-EM structures reveal that central guide–target pairing pries open AGO2’s central cleft, driving AGO2 into the preactive state required for target cleavage.

A pervasive RT–qPCR artifact inflates RNA knockdown by RNA-targeting CRISPR -
Yikey Yikes! BTW, similar concerns apply to siRNAs/ASOs that are sometimes suspected to affect RT and/or PCR . So nice somebody looked into this! rdcu.be/RUYFaGa649va

I’m thrilled to share my latest research with @claudiakohler11.bsky.social in @mpi-mp-potsdam.bsky.social, now published in @natplants.nature.com ! In this work, we reveal that long-distance transported siRNAs from maternal sporophytic tissues are functional in pollen development and reproduction.

Things I learned in grad school:
- How to use siRNAs for targeted gene suppression
- Protocols for Northern, Southern, & Western blots

Things I did not learn in grad school:
- How to run a lab when the government is falling apart
- How to get funding for "woke science"
