#ACOD1
They also measured a respirometer to compare normal zebrafish vs transgenic zebrafish with ACOD1 and found that these ACOD1 fish produced less energy, consuming less oxygen, and moving less than their counterparts.

A new #ScienceAdvances study identifies a neuroimmune pathway for treating sepsis, finding that dopamine inhibits expression of a critical regulator of immunometabolism.

This suggests that pramipexole, a dopamine agonist, could effectively treat lethal infections. scim.ag/4kfAfrZ

CLYBL and ACOD1 are the two enzymes which govern the process; Phair poses that the ratio of these two (CLYBL: ACOD1) will govern your disease baseline.

3) Hard evidence linking this proposed mechanism to ME/CFS is lacking. The ACOD1 gene, which plays a key role in the theory, didn’t show a signal in the big DecodeME study.

So, could ACOD1 expression be inhibited and reduce fx?

They tested four compounds that might inhibit ACOD1 and found that there was evidence towards resumed function and increased locomotion in the zebrafish in question.

Do you know if ACOD1 was significant in DecodeME?

6) It's unclear, however, if this model fits ME/CFS. At high expression of ACOD1, the fish also showed a reduced heart rate and signs of cardiac impairment.

7) Lastly, they tested multiple ACOD1 inhibitors and found that some restored movement in the zebrafish back to a normal amount. So if the Itaconate shunt model is correct, this could be a potential treatment.

2) The hypothesis states that the normal TCA cycle to generate energy is disrupted during an infection. A gene called ACOD1 diverts the metabolites away from the cycle towards the Itaconate pathway, making it less efficient in producing energy.

4) In tests with lights or vibration as stimuli being turned on and off, the zebrafish with ACOD1 overexpression had approximately a 40% reduction in movement compared to controls.

They also had reduced oxygen consumption.

5) The gene expression was regulated using heat-shock so the researchers could test different versions of ACOD1 overexpression by increasing the temperature of the water.

The more the gene was expressed, the greater the signs of energy deficits.
