Really excited to have our work out in the world. CCHFV has a 450kDa L-Protein, about twice the size of L-Proteins from other segmented RNA viruses. We have used cryoEM and functional assays to begin to understand it this massive multifunctional protein works.
Structural and functional characterisation of the Crimean-Congo Haemorrhagic Fever Virus RNA Dependent RNA Polymerase
Crimean-Congo Haemorrhagic Fever Virus (CCHFV) is found across Africa, Asia, and the Middle East where it can cause Haemorrhagic outbreaks with high case fatality rates. Central to the viral life cycl...

Structures and inhibition of the Crimean–Congo haemorrhagic fever virus polymerase - Nature
Structural studies of Crimean–Congo haemorrhagic fever virus (CCHFV) polymerase reveal the structural basis of CCHFV RNA synthesis and highlight druggable sites for designing both nucleoside antivirals and non-nucleoside antivirals.

Cryo-EM structures of CCHFV polymerase reveal a stepwise initiation stabilization pathway and a dual-site inhibition mechanism
Yang et al. report cryo-EM structures of the CCHFV RNA polymerase across multiple functional states. These structures reveal a promoter-dependent initiation stabilization pathway and a dual-site mechanism of suramin inhibition, providing insights into bunyavirus polymerase regulation and inhibition.

In addition, we imaged rVSVs displaying ANDV and CCHFV glycoproteins (provided by @chandranlab.bsky.social) and worked with @jesseerasmus.bsky.social and colleagues at HDT Bio to assess the VLPs' immunogenicity in mice via repRNA. Thanks to all authors for their contributions!

The trend with tuberculosis was pretty prediction considering that we know how synergistic it with the immune dysfunction from HIV so it would likely behave the same with SARS-CoV-2.

Also, CCHFV is the dark horse for vector borne diseases right now as it also has airborne transmission.

16/24 are viral… 14 RNA… (8 neg sense…) 12 zoonotic… 5 arboviruses… 9 mucosal…

One way to fix it would be group the bunyaviruses together (lassa, hanta, cchfv, oropouche, rift)
www.bbc.co.uk/news/article... a nice step forward but annoyingly inconsistent as some “pathogens” are species (eg Lassa) and others are whole orders of viruses (Flaviviridae). I think there’s better way to focus people. Interesting are those not on it though.
Priority pathogens: UK draws up new disease-threat watch list
Some are viruses with global pandemic potential - like Covid - others infectious illnesses with no treatments.
