#Cytonemes
Great two days at our Wnt SFB 1324 meeting @heidelberg ! 🧬

Fantastic science, fresh ideas, new tools & lots of exciting collaborations. Great to have Lizzy, Gemma & Jess presenting their work and getting inspired.

Now back @lsiexeter.bsky.social to try it all out! 🔬🐟 #Wnt #Cytonemes

How does a Wnt cytoneme contact become a productive signalling site?

Emma Cooper and colleagues find focal Ca²⁺ transients at cytoneme contacts and link Ca²⁺ availability to contact stability and Wnt/β-catenin signalling.

What if we didn’t use genetics (says the fly scientist) but used other, physical, manipulations? What if we could put up molecular jersey barriers to keep cytonemes from interacting or moving normally? Would it lead to tissue perturbations?

One of the more annoying things about this system is that genetic disruptions of actin cytoskeleton that mess with cytonemes also disrupts events across the whole cell - which is you know, not great if your signaling system requires say, actin-dependent endocytosis (Notch)

Wouldn’t it be cool if we had a more specific way to disrupt cytonemes? (flies don’t have MyoX 🙃 )

So we embedded dissected patterning tissues into a calcium-inducible nanofiber gel and asked what happened to their cytonemes, and what happened to their patterning.

Our lab is interested in cell shape, differentiation and tissue patterning. We have been using cytonemes and Notch signaling in the patterning peripheral nervous system of the fly as a model system. We have a lot of questions, here’s one of my favorite addressed in this paper:

First, their cytonemes get messed up. And they don’t move very well. This was possibly one of the most difficult experiments I’ve ever asked someone to do and my PhD student did a stellar job.

Faisal Mahdi's review made the cover🎉 what a great end to a long first week of the semester!

Aiyana Lopez, a NERSC USRA recipient, did some incredible work investigating how the Notch protein regulates cytonemes and, conversely, how cytonemes regulate Notch signaling!

Last, but definitely not least, is our fantastic returning co-op student, Christina Bernados. She investigated the role of cytonemes in lung health and developed machine-learning pipelines to automate cytoneme quantification, something we will be using in the lab for years to come!

New review out!🎉🧪🔬 Cancer cells don't just broadcast signals, but deliver them directly. TNTs and TMs get most of the hype, but our review explores how cytonemes are reshaping our understanding of tumor communication and the TME.

Happy #MicroscopyMonday! Here's some #SciArt of a confocal micrograph stylized using the Ps emboss filter. See the thin projections extending between the 2 cells? They're #cytonemes! 🔬 How do they find each other? Follow postdoc @itsscientistsam.bsky.social to learn more 🧪 👩‍🔬

Well - I’m not sure. It’s the mTmG lineage tracer, so the labeled cells expressed Shh at one point during development, but may not be expressing it at this stage. We’re trying to ID cell types that might make SHH cytonemes during development - currently in the “survey” stage. 🤓
