The wwPDB just hit a major milestone: 𝟏𝟎,𝟎𝟎𝟎 𝐄𝐌𝐃𝐁 𝐞𝐧𝐭𝐫𝐢𝐞𝐬 𝐫𝐞𝐥𝐞𝐚𝐬𝐞𝐝 𝐢𝐧 𝐚 𝐬𝐢𝐧𝐠𝐥𝐞 𝐲𝐞𝐚𝐫. To put this in context, the entire EMDB reached 10,000 entries total in 2019 — now the archive 𝐡𝐚𝐬 𝐬𝐮𝐫𝐩𝐚𝐬𝐬𝐞𝐝 𝟓𝟎,𝟎𝟎𝟎. Huge credit to the community driving 3DEM forward!

📄 New paper: 3DEM Structure Map Validation Recommendations

Two new metrics assess model-to-map fitness in cryo-EM:
🔹 Q_relative_all → compares avg Q-score across PDB/EMDB
🔹 Q_relative_resolution → compares within similar resolution

With 8 composite cryo-EM maps (totaling 47 EMDB entries) and several biochemical assays, we demonstrate the mechanistic basis for the positioning of H3K36me3 within actively transcribed regions. Surprisingly, SETD2 must be relieved of auto inhibition by the transcription machinery!
Ever wondered how transcription choreographs histone modifications? Our work reveals the basis of co-transcriptional H3K36me3 by SETD2. We visualize how a histone writer coordinates with the transcription machinery! This is the magnus opus of @jonmarkert.bsky.social!

Now on @biorxivpreprint.bsky.social: Dari Kimanius' paper on #Blush regularisation, a machine-learning based prior in #RELION5 trained on EMDB #cryoEM half-maps, which he uses to solve a #cryoEM data set of a 40 kDa protein:RNA complex that was previously intractable!

Apparently non-existent density for a ligand in PDB 7yjj and EMDB 33872 is up for interpretation by readers and does not affect the robustness of the authors' paper......

This is a fantastic example of how to begin as a newcomer to #cryoEM. Being proactive in getting advice from experienced scientists and going to any of the great workshops would likely lower the incidences of erroneous structures in the EMDB and PDB (There’s not a lot but there are a few).

This week's release of 200 entries by the EMDB can be viewed at www.ebi.ac.uk/emdb. The running total for the month of May is 650 entries bringing the archive to a total of 57595 entries.
