Jiatong Li et al. reveal that glutamine metabolic reprogramming drives pancreatic cancer resistance to KRASG12D inhibitors via ANXA1-mediated stabilization of GOT1 and mitochondrial translocation of GLS1. Glutamine restriction, either dietary or pharmacological, overcomes this resistance, offering a promising therapeutic strategy.
