Glutaredoxins (Grx) rapidly reduce glutathione polysulfides (GSSnSG) to glutathione hydroper/polysulfides (GSSnH) or GSSnH to hydrogen (poly)sulfides (H2Sn). Reduction of glutathionylated glutaredoxins by reduced glutathione (GSH) yields glutathione disulfide (GSSG). Thus, steady-state concentrations of glutathione (hydro)per/polysulfides should be tightly controlled in compartments containing active glutaredoxins and high GSH concentrations. ABSTRACT Hydropersulfides have gained attention as excellent biochemical nucleophiles and membrane-protective radical scavengers. They form perthiyl radicals, which terminate radical chain reactions through self-recombination, yielding polysulfides. It is currently unknown exactly how polysulfides are subsequently reduced. Using stopped–flow kinetic measurements in combination with mass spectrometry, we show that the model class I glutaredoxin from Plasmodium falciparum (PfGrx) rapidly reduces the polysulfides glutathione trisulfide (GS3G) and tetrasulfide (GS4G), yielding glutathionylated PfGrx and the corresponding glutathione hydropersulfide (GSSH) and hydrotrisulfide (GS3H). The second-order rate constants of these reductions ≥107 M−1s−1 are even slightly higher than for glutathione disulfide (GSSG). In contrast, PfGrx was inactive or only moderately active toward cystine or cysteine trisulfide. GSSH and GS3H are further reduced by PfGrx with second-order rate constants on the order of 106−107 M−1s−1, yielding glutathionylated PfGrx and hydrogen sulfide (H2S) or hydrogen disulfide (H2S2), respectively. Thus, glutaredoxins specifically recognize the glutathione moiety of glutathione (hydro)per/polysulfides. Since reduced glutathione (GSH) rapidly reduces glutathionylated glutaredoxins, glutathione (hydro)per/polysulfides are efficiently converted to GSSG and H2S or the corresponding hydrogen polysulfides. As a consequence, the steady-state concentration of glutathione (hydro)per/polysulfides should be tightly controlled in subcellular compartments containing active glutaredoxins and high GSH concentrations.

We sought to evaluate oxidative changes in premature infants receiving 100% oxygen compared with 30% during deferred cord clamping (DCC). Methods Premature infants born at 220/7 to 286/7 weeks received DCC in conjunction with either 30% (LO Group) or 100% (HI Group) oxygen. Blood was extracted from a preserved umbilical segment and a postnatal sample was collected from umbilical vascular lines within two hours of birth. Reduced-to-oxidized glutathione (GSH/GSSG) ratios were analyzed using liquid chromatography coupled to tandem mass spectrometry. Results Sixty-eight infants had data available for analysis. The median (IQR) gestational age of infants was 264/7 (246/7, 282/7) weeks in both groups. Among infants receiving 100% versus 30% oxygen, median (IQR) GSH/GSSG ratio were not statistically different in arterial cord blood [7.5 (0.6, 290) vs 37 (1.1, 265), p = 0.52] or venous cord blood [8.4 (2,50) vs 76 (5, 210), p = 0.12] or postnatal samples [14 (2, 290) vs 8 (2, 280), p = 0.98)]. Conclusion Briefly providing 30% vs. 100% oxygen for 90 seconds during DCC showed no significant difference in GSH/GSSG ratios, but redox effects remain unclear given variability, sample size and limited power. The endoplasmic reticulum (ER) requires an oxidative environment to support the efficient maturation of secretory and membrane proteins. This is in part established by glutathione, a redox-active metabolite present in reduced (GSH) and oxidized (GSSG) forms. The ER maintains a higher GSSG:GSH ratio than the cytosol; however, the mechanisms controlling ER redox balance remain poorly understood.
