Systemic autoinflammatory diseases (SAID) with inborn errors of cell death (IECD) are caused by overactivation of programmed cell death (PCD). However, the pathogenesis by which PCD leads to autoinflammation remains unclear. Here, we identified IECD patients carrying compound heterozygous RIPK1 variants K377E/R390G with autoinflammatory manifestations. Mechanistically, K377E and R390G mutations suppress NF-κB signaling and activate RIPK1 to promote cell death. CD8+ T cells of the patients displayed overactivated RIPK1 and excessive cell death, leading to an elevated CD4/CD8 ratio, which could also be detected in patients with cleavage-resistant mutation of RIPK1 or SHARPIN deficiency. We show that the increased cell death of CD8+ T cells promotes TNF and IFN-γ secretion to activate monocytes/macrophages, which triggers overproduction of proinflammatory cytokines. Staff Reporter ONLY about 28% of Grade 3 learners met the minimum literacy pass rate, while just 32% passed in numeracy in 2024, according to the National End-of-Phase Grade 3 Literacy and Numeracy Assessment. This was highlighted by the Minister of Education, Innovation, Youth, Sport, Arts and Culture, Sanet Steenkamp, at the recent relaunch of the RightStart Programme. This programme aims to provide integrated early childhood development (IECD) for all children from birth to age eight. “Current data reveals a sobering reality: nine out of ten African learners aged 10 are unable to read a simple text or perform basic maths tasks – this is what we define as Learning Poverty. In Namibia, our 2024 National End-of-Phase Grade 3 Literacy and Numeracy Assessment showed that only 28% of learners achieved the minimum pass rate in literacy, and only 32% in numeracy. Photos: Ministry of Gender Equality and Child Welfare. --- She acknowledged that there are critical obstacles within the country’s Early Childhood Development (ECD) sector, including a shortage of qualified educators, which exacerbates learning poverty. “Of 414,069 children aged 0–4, only 51,872 accessed ECD services in 2024, meaning over 87% of children aged 0–4 do not attend integrated ECD programmes, and 60% of pre-primary-aged children are not enrolled in preschool. Only 17% of educators hold formal qualifications. These represent significant barriers to lifelong learning and contribute directly to learning poverty,” Steenkamp said. She assured that the government understands the importance of addressing these challenges, which is what inspired the relaunch of the RightStart Programme. The minister explained that the programme’s goal is to reduce learning poverty and strengthen foundational learning outcomes by delivering accurate, practical, and culturally relevant information to parents, caregivers, teachers, and communities. “The RightStart Programme plays a central role in our national strategy to address learning poverty and improve foundational learning outcomes. It places the child at the heart of development, beginning from pregnancy through to age eight, while also empowering parents, caregivers, teachers, and leaders with clear, actionable information. What makes this initiative powerful is its focus on practical change: shifting knowledge, reshaping attitudes, and transforming daily practices to elevate early childhood development from a peripheral concern to a national development priority,” Steenkamp explained. The programme will be implemented by the Ministry of Education, Innovation, Youth, Sport, Arts and Culture, the Ministry of Health and Social Services, the European Union, InterTeam, and UNICEF. “RightStart is positioned to promote community engagement and demand generation through national awareness campaigns. We are committed to leveraging the RightStart Campaign for National Mobilisation, expanding it to all 14 regions, embedding its materials into community programmes, and collaborating with media and influencers. Our targets reflect this ambition: for the campaign to be operational in all 14 regions, for 80% of parents to report increased awareness, and for 100% of regional plans to incorporate RightStart activities,” Steenkamp added.
